E-ROOTREE - Internship Opportunity
This is a short-term grant and the work would be immediate [completed before May end] and flexible. Paid as per your recommendation and study is extended as per the results we obtain. This study is not for academic purposes but for product design and engineering feasibility evaluation.
1. Plant Biology and Physiology
- Understanding Plant Defense Mechanisms: Knowledge of plant responses to wounding, including callose deposition, Tylosis, reactive oxygen species (ROS) production, and hormonal signaling pathways like jasmonic acid and salicylic acid.
- Plant Tissue Anatomy: Familiarity with xylem, phloem, and other tissues to identify structural changes and locate defense-related features.
2. Microscopy Techniques
- Confocal Microscopy Proficiency: Expertise in operating confocal microscopes, including setting laser parameters, adjusting pinhole size, and performing Z-stack imaging for 3D reconstructions.
- Fluorescence Microscopy: Knowledge of fluorescence principles, selecting appropriate excitation/emission wavelengths, and handling photobleaching issues.
3. Staining and Imaging
- Dye Handling: Familiarity with fluorescent dyes like Aniline Blue, DCFH-DA, or Alexa Fluor dyes, and the ability to optimize staining protocols for specificity.
- Sample Preparation: Skills in tissue fixation, sectioning, clearing, and mounting for confocal imaging.
4. Molecular Biology Techniques
- Hormone and ROS Analysis: Capability to interpret the molecular roles of plant hormones and oxidative stress during wounding.
5. Image Processing and Data Analysis
- Software Expertise: Proficiency in using image analysis tools like FIJI (ImageJ), Imaris, or Zeiss Zen for quantifying fluorescence intensity, co-localization, and 3D reconstructions.
- Statistical Analysis: Ability to analyze and interpret experimental data, including comparisons across different treatments or conditions.
6. Experimental Design and Execution
- Designing Experiments: Knowledge of control setups, replicates, and randomization for reliable results.
7. Collaboration Skills
- Working with multidisciplinary teams, including biologists, pathologists, and imaging specialists, to design and execute experiments effectively.
